Abstract
Phase equil. behavior of antibiotics is important in drug design and for optimization of the recovery process in manufg. Aq./org. partitioning behavior of a clin. important antibiotic, oxytetracycline was measured. The org. phase included pure solvents such as n-hexane, CHCl3, di-Et ether and EtOAc. In addn., measurements were also made for mixed org. phase composed of n-hexane + EtOAc. Org.-phase partitioning increased with the H-bonding tendency of the solvent in the order n-hexane <CHCl3 <di-Et ether <EtOAc. Single solvent-org.-phase data were correlated using a previously described activity coeff. model to obtain the free-energy-of-H-bonding parameters for H-bonding between the antibiotic and the solvent. Predictions made for mixed-solvent-org.-phase systems agreed well with the exptl. data without using any adjustable parameter.
