My advice on the matters for the day (as listed in the calendar):

  • Meselson & Stahl (1958): If you haven't yet figured out how the Meselson & Stahl experiment works, now is the time.
     
  • Entire semester.: Think back on the problem sets, study questions, and tutorials that have appeared thus far... Where do you need help?
     
  • Problem Set 4: Problem 6 is going to be the main event. If you haven't done so already, I'd advise taking the time to understand the basics of PCR (e.g. through one of the videos).

    Anything further you'd like to discuss regarding Meselson and Stahl (1958) and its companion?
    In particular, do you understand dispersive DNA replication (Fig. 3 of the companion)?
    Do you understand how to read Fig. 4 of Meselson and Stahl (1958)?
    Do you understand the experiment underlying that figure?
    Can you recreate Fig. 4 based on different views of reality (e.g. DNA replicates conservatively)?

    Anything further you'd like to discuss regarding one or more problems in this problem set?
    Do you have a high level of comfort concerning the role of primers in PCR (enough so that you could figure out primers yourself)?
