My advice on the matters for the day (as listed in the calendar):
I. Basic Information
A. Your name
II. Old business
- Gene Regulation and Problem Set 7 (questions 5 and 6)
Regarding the notes on Gene Regulation and Bacteriophage and the two related problems (5 and 6) in Problem Set 7:
- Any problems relating the promoter and operator sites in the Lambda cI-cro region to consensus sequences?
- Any problems finding genes in TP901-1 that may be analogous to cI and cro and possible regulatory sites between them?
- Are you OK with higher order issues, like how cI and Cro proteins work in relation to RNA polymerase?
- Or even higher than that, how regulatory feedback loops work to make stable decisions regarding lysogeny vs lysis?
- Or higher or lower or sideways... anything else?
- Problem Set 7, questions 1 through 4)
Any concerns about other questions in
Problem Set 7?
- Semester Project
Drawing on either:
are you feeling some confidence that you can put together an award winning proposal or translation? Anything you need? (besides time) (and if you need time, see the last line of advice here). Anything you'd like to discuss?
III. Belasco et al (1985), Part I
A. How far have you gotten in this article and its
companion (Part I)?
B. Consider how comfortable you are with the following topics regarding these matters:
- The question you are directing at this article and the biology behind the question
- The nature of an operon
- The process of skimming an article to decide what part of it to read
- S1 protection experiments
IV. Belasco et al (1985), Part II (if you care to sample it)
A. How far have you gotten in this article and its
companion (Part II)?
B. Consider how comfortable you are with the following topics regarding these matters:
- The elements of the S1-protection experiment in this article
- Use of BioBIKE to investigate genome of R. capsulatus
- Equating Belasco et al's Fig. 1 with the genome sequence obtained from BioBIKE
- Identification of restriction sites (e.g. EcoRI)
- Interpreting Fig. 2 from Belasco et al
- Interpreting Fig. 4 from Belasco et al
